Background:

Bovine tuberculosis (bTB) causes significant global livestock losses, persisting despite eradication efforts.
Current diagnostic methods (skin tests, interferon-γ assays) have limitations in sensitivity and specificity, hindering complete eradication.
Early and reliable diagnostics are essential to manage bTB, especially given its reemergence from wildlife reservoirs.

Purpose of the Study:

To identify a comprehensive panel of immune response genes associated with bovine tuberculosis (bTB) using high-throughput sequencing.
To develop and evaluate a reliable bTB classification framework based on RNA-Seq data from peripheral blood mononuclear cells (PBMCs).

Main Methods:

High-throughput sequencing of PBMCs transcriptome from healthy and bTB-infected cattle at different disease stages.
Splice-alignment of over 90 million RNA-Seq reads against the reference genome.
Differential gene expression analysis to identify genes responding to bTB infection.

Identified 2,312 differentially expressed genes in response to bTB infection (p<10(-8)).
Achieved over 99% accuracy in classifying bTB infected status using split-sample validation.
