Summary
A novel bacterium, Methylobacterium tarhaniae sp. nov., was identified from Nigerian arid soil. This facultative methylotroph expands the known diversity within the Methylobacterium genus.
Area of Science:
- Microbiology
- Bacteriology
- Environmental Microbiology
Background:
- The genus Methylobacterium comprises diverse Gram-negative bacteria known for their metabolic versatility.
- Facultative methylotrophic bacteria play significant roles in various ecosystems, including soil environments.
Purpose of the Study:
- To characterize a novel bacterial isolate from arid soil in Abuja, Nigeria.
- To determine the taxonomic position of the isolate using a polyphasic approach.
Main Methods:
- Polyphasic taxonomic analysis including 16S rRNA gene sequencing.
- Phylogenetic analysis, DNA-DNA hybridization, and chemotaxonomic analyses (fatty acids, ubiquinones).
- Determination of DNA G+C content.
Main Results:
- Strain N4211(T) was identified as a reddish-orange-pigmented, Gram-negative, aerobic, facultative methylotroph.
- Phylogenetic analysis placed strain N4211(T) within the genus Methylobacterium, closely related to M. aquaticum, M. platani, and M. variabile.
- DNA-DNA relatedness values (60.0% with M. aquaticum, 48.2% with M. platani) and distinct chemotaxonomic profiles supported its novelty.
- The DNA G+C content was determined to be 67.3 mol%.
Conclusions:
- Strain N4211(T) represents a new species within the genus Methylobacterium.
- The proposed name for this novel species is Methylobacterium tarhaniae sp. nov.
- The type strain is N4211(T) (= KCTC 23615(T) = DSM 25844(T)).

Bacterial Phylum Tenericutes
The phylum Tenericutes, which includes the single class Mollicutes, comprises bacteria that lack cell walls. The term "Mollicutes" derives from the Latin word mollis, meaning "soft." These organisms are among the smallest known and are commonly referred to as mycoplasmas due to the prominence of the genus Mycoplasma, which includes well-known human pathogens. Despite their inability to stain gram-positively (a result of their lack of cell walls), mycoplasmas are phylogenetically related to the...
Bacterial Phylum Actinobacteria
Coryneform bacteria are gram-positive, aerobic, nonmotile rods that exhibit irregular, club-shaped, or V-shaped arrangements. Their V-shape results from snapping division, where the inner cell wall layer forms the cross-wall, while the outer layer remains intact until it ruptures on one side, causing the daughter cells to bend away. The primary genera are Corynebacterium and Arthrobacter. Corynebacterium includes diverse species, ranging from saprophytes to pathogens like Corynebacterium...
Hyperthermophilic Bacteria
Domain Bacteria includes some unique hyperthermophilic species. They exhibit remarkable adaptations that enable survival in extreme environments.Thermotoga species are rod-shaped, gram-negative, non-sporulating hyperthermophiles that form a sheath-like envelope called a toga. They ferment sugars or starch, producing lactate, acetate, CO₂, and H₂, and can also grow via anaerobic respiration using H₂ and ferric iron. Found in hot springs and hydrothermal vents, over 20% of their genes show strong...
Microbes and Methanogenesis
Methanogenesis is a critical microbial process in anaerobic ecosystems responsible for the biological production of methane, a potent greenhouse gas and valuable biofuel. This metabolic pathway is primarily facilitated by methanogenic archaea, which thrive in anoxic environments such as wetlands, sediments, and animal gastrointestinal tracts. The absence of oxygen in these habitats prevents aerobic respiration, thereby favoring alternative biochemical pathways for organic matter degradation.In...
Anoxygenic Phototrophic Bacteria
Anoxygenic phototrophic bacteria are a diverse group of microorganisms that perform photosynthesis without producing oxygen. They primarily include purple sulfur bacteria, purple nonsulfur bacteria, green sulfur bacteria, and green nonsulfur bacteria. These bacteria are classified into the Gammaproteobacteria, Alphaproteobacteria, Betaproteobacteria, Chlorobi, and Chloroflexi lineages, each with distinct physiological and ecological adaptations.Purple sulfur bacteria belong to the...
Other Unique Bacteria
Magnetic bacteria exhibit a directed movement called magnetotaxis, driven by structures called magnetosomes. These magnetosomes consist of chains of magnetic particles made of either magnetite (Fe₃O₄) or greigite (Fe₃S₄) and are organized in a linear conformation by a protein scaffold within invaginations of the cell membrane. The bacteria align along the north–south magnetic field lines, much like a compass needle. They are typically microaerophilic or anaerobic and are commonly found near the...
