Interleukin enhancer binding factor 3 (Ilf3) and Nuclear Factor 90 (NF90) are RNA binding proteins with unclear functions. This study identified six new protein partners, suggesting Ilf3 and NF90 play roles in mRNA metabolism.
Area of Science:
- Molecular Biology
- Cell Biology
- Biochemistry
Background:
- Interleukin enhancer binding factor 3 (Ilf3) and Nuclear Factor 90 (NF90) are proteins derived from the ILF3 gene.
- Their precise functions are not fully understood, despite being known as RNA binding proteins involved in various cellular processes.
- Their roles are influenced by interacting partners, protein complexes, and cellular location.
Purpose of the Study:
- To investigate the protein partners of Ilf3 and NF90.
- To elucidate the functions of Ilf3 and NF90 in cellular phenomena.
- To establish a link between Ilf3/NF90 functions and their interacting proteins.
Main Methods:
- Affinity chromatography was employed to identify protein partners.
- Analysis of protein interactions with double-stranded RNA binding motifs.
- Characterization of protein complexes involving Ilf3 and NF90.
Main Results:
- Six novel protein partners interacting with Ilf3 and NF90 were identified: hnRNP A/B, hnRNP A2/B1, hnRNP A3, hnRNP D, hnRNP Q, and PSF.
- These hnRNP proteins are involved in mRNA stabilization, transport, and translation regulation.
- PSF was identified as a splicing factor.
- Ilf3, NF90, and most identified partners exist in large molecular complexes.
Conclusions:
- The identified partners and their known functions suggest a role for Ilf3 and NF90 in mRNA metabolism.
- This study links the RNA binding capabilities of Ilf3 and NF90 to the functions of their interacting partners.
- Further research into these interactions can clarify the complex cellular roles of Ilf3 and NF90.
