Summary

The PowerPlex® Y23 System enhances Y-STR analysis by adding six new highly discriminating loci, improving individual distinction. This robust kit reliably amplifies DNA from various sources, even with inhibitors and mixed DNA samples.

Background:

  • Current Y-STR kits have limitations in discriminating between unrelated males.
  • There is a need for enhanced Y-STR analysis systems with higher discriminatory power.

Purpose of the Study:

  • To introduce and validate the PowerPlex® Y23 System, a novel Y-STR amplification kit.
  • To assess the system's performance in discriminating individuals and handling challenging samples.

Main Methods:

  • The PowerPlex® Y23 System combines 17 established Y-STR loci with 6 new, highly discriminating loci.
  • Amplification from purified DNA extracts and direct amplification from forensic substrates were performed.
  • Validation studies included FBI and SWGDAM requirements, inhibitor tolerance tests, and sensitivity assessments.

Main Results:

  • The six new Y-STR loci exhibit higher gene diversities, enhancing individual discrimination.
  • The system demonstrated robustness against common inhibitors (hematin, humic acid, tannic acid).
