Summary

Technetium-99m labeled (99m)Tc-(Me)FGCDEVD preferentially accumulates in apoptotic cells. This novel tracer shows potential for detecting apoptosis in vivo, particularly after chemotherapy-induced cell death.

Area of Science:

  • Biomedical Imaging
  • Radiochemistry
  • Molecular Imaging

Background:

  • Apoptosis, or programmed cell death, is crucial in development and disease.
  • Detecting apoptosis in vivo is vital for monitoring treatment response and disease progression.
  • Current methods for apoptosis detection have limitations in sensitivity and specificity.

Purpose of the Study:

  • To develop and evaluate a novel technetium-99m labeled tracer for apoptosis detection.
  • To assess the in vitro and in vivo performance of (99m)Tc-(Me)FGCDEVD for imaging apoptosis.

Main Methods:

  • Radiolabeling of (Me)FGC(Bz)DEVD with technetium-99m (99mTc) to achieve high yield.
  • In vitro studies to evaluate tracer accumulation in apoptotic cells.
  • In vivo studies in tumor-bearing models to assess tumor uptake after cisplatin-induced apoptosis.

Main Results:

  • High-yield radiolabeling of (Me)FGC(Bz)DEVD with 99mTc was achieved.
  • (99m)Tc-(Me)FGCDEVD demonstrated preferential accumulation in apoptotic cells in vitro.
  • Significant tumor uptake of the tracer was observed in vivo following cisplatin administration, correlating with apoptosis induction, but not in untreated tumors.

Conclusions:

  • (99m)Tc-(Me)FGCDEVD is a promising radiotracer for detecting apoptosis.
  • The tracer's ability to visualize chemotherapy-induced apoptosis in vivo offers potential for clinical applications in cancer therapy monitoring.
