A new method, multiple acceptors PbFRET (Ma-PbFRET), accurately measures Förster resonance energy transfer (FRET) efficiency in constructs with multiple acceptors. This technique is valuable for studying large biomolecular complex assembly in living cells.

Area of Science:

  • Biophysics
  • Molecular Biology
  • Biochemistry

Background:

  • Partial photobleaching FRET (PbFRET) is a simple method for measuring FRET efficiency.
  • Existing PbFRET methods cannot analyze constructs with multiple acceptors, limiting their application for larger Förster distances.

Purpose of the Study:

  • To develop a novel method for quantifying FRET efficiency in constructs with multiple acceptors.
  • To extend the applicability of PbFRET to complex biomolecular systems.

Main Methods:

  • Developed a linear theory for acceptor photobleaching probability in multi-acceptor systems.
  • Created the multiple acceptors PbFRET (Ma-PbFRET) method to measure FRET efficiency.
  • Validated Ma-PbFRET in living cells using constructs with two or three acceptors.

Main Results:

  • Ma-PbFRET accurately quantifies FRET efficiency in constructs with multiple acceptors.
  • The method is effective across varying degrees of acceptor photobleaching.
  • Experimental validation confirmed the method's precision in living cells.

Conclusions:

  • Ma-PbFRET provides a simple and powerful tool for analyzing biomolecular complex assembly and disassembly.
  • Enables investigation of molecular interactions over larger distances in vivo.
  • Overcomes limitations of previous FRET methods for multi-acceptor systems.
