Methods to Assess Microbial Populations01:30

Methods to Assess Microbial Populations

Assessing microbial populations is crucial for understanding microbial roles in health, ecology, and industry. Various complementary techniques—both culture-based and molecular—enable detailed analysis of microbial abundance, diversity, and function.Viable Plate CountThe viable plate count is a traditional culture-based method used to estimate the number of living microbes in a sample. After serial dilution, the sample is spread onto nutrient agar plates. Each viable cell forms a visible...
Fixation and Sectioning01:03

Fixation and Sectioning

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens.
The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Solid specimens, such as skin scraping, can be placed on the slide before adding a drop of liquid to prepare the wet mount. Sometimes the liquid is simply water, but stains are often added...

Area of Science:

  • Biotechnology
  • Molecular Diagnostics
  • Biochip Technology

Background:

  • Conventional cell staining methods lack specificity and sensitivity.
  • Antibodies, while common, can be expensive and difficult to handle.
  • Emerging biochip technology offers potential for improved cell detection.

Purpose of the Study:

  • To develop and evaluate aptamer-based biochips for selective cell detection and quantitation.
  • To compare the performance of aptamers with antibodies as ligands on biochips.
  • To assess the suitability of aptamers for future diagnostic development.

Main Methods:

  • Immobilization of single-stranded RNA aptamers specific for HER2 or PSMA on biochips.
  • Utilizing HER2- or PSMA-positive cells as analytes in a model system.
  • Combination of biochip technology with aptamer-based recognition.

Main Results:

  • Aptamers demonstrated equivalent or superior specificity and sensitivity compared to antibodies.
  • The PSMA-specific aptamer sensor exhibited excellent stability, reproducibility, and reusability.
  • A detection limit as low as 10^3 LNCaP cells was achieved with the PSMA sensor.

Conclusions:

  • Nucleic acid aptamers are suitable low molecular weight receptors for biochips.
  • Aptamer-based biochips enable sensitive and specific cell detection and quantitation.
  • This technology holds significant potential for the development of future diagnostic tools.
