Summary

Zebrafish dynamin-2 (DNM2) genes are essential for development. Studying these zebrafish DNM2 orthologs provides insights into human neuromuscular disorders like centronuclear myopathy and Charcot-Marie-Tooth disease.

Area of Science:

  • Molecular Biology
  • Developmental Biology
  • Genetics

Background:

  • Dynamin-2 (DNM2) is a GTPase crucial for endocytosis and cellular trafficking.
  • Mutations in human DNM2 are linked to centronuclear myopathy and Charcot-Marie-Tooth disease.
  • Zebrafish serve as a valuable model for studying neurological disorders.

Purpose of the Study:

  • To characterize the zebrafish dynamin-2 (dnm2 and dnm2-like) orthologs.
  • To investigate the role of zebrafish DNM2 in development.
  • To assess the potential of zebrafish as a model for DNM2-related human diseases.

Main Methods:

  • Gene and protein structure analysis of zebrafish dnm2 and dnm2-like.
  • Expression pattern analysis during zebrafish development and in adult tissues.

  • Knockdown studies using morpholinos and rescue experiments with human DNM2 RNA.
  • Main Results:

    • Zebrafish dnm2 and dnm2-like genes show structural similarity to human DNM2.
    • Both genes are expressed throughout zebrafish development and in adult tissues.
    • Knockdown of dnm2 and dnm2-like caused significant developmental abnormalities, which were rescued by human DNM2 RNA.

    Conclusions:

    • Zebrafish dnm2 and dnm2-like are functional orthologs of human DNM2.
    • These genes are essential for normal zebrafish development.
    • The study validates zebrafish as a relevant model for understanding DNM2-related neuromuscular disorders.
