Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal01:22

Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal

Erythropoietin-producing hepatocellular carcinoma receptor (Eph) and its ligand, Eph receptor-interacting protein (Ephrin) were first discovered in the human carcinoma cell line, hence the name. Ephrin-Eph interaction guides cells to reach their appropriate location in adult tissues. They also play an essential role in the immune system by helping in immune cell migration, adhesion, and activation. Based on their structure and function, Eph is divided into two classes — EphA and EphB.

Summary
This summary is machine-generated.

Human eosinophils express the CCR7 receptor, enabling them to respond to chemokines like CCL19 and CCL21. This finding is crucial for understanding eosinophil migration in allergic inflammation.

Area of Science:

  • Immunology
  • Cell Biology
  • Molecular Biology

Background:

  • Human eosinophils are key players in allergic inflammation.
  • Eosinophil migration to lymph nodes is observed, but the underlying mechanisms are unclear.
  • CCR7 is a critical chemokine receptor for leukocyte trafficking to lymph nodes.

Purpose of the Study:

  • To investigate CCR7 expression in human eosinophils.
  • To determine functional responses of eosinophils to CCR7 ligands CCL19 and CCL21.

Main Methods:

  • Purification of human eosinophils from healthy donors.
  • Assessment of CCR7 expression using flow cytometry and Western blot.
  • Chemotaxis, shape change, calcium flux, and ERK phosphorylation assays.

Main Results:

  • Human eosinophils express CCR7 on their surface.
  • IL-5-primed eosinophils show dose-dependent chemotaxis to CCL19 and CCL21.
  • Eosinophils exhibit shape changes, calcium influxes, and ERK phosphorylation upon stimulation with CCL19/CCL21.

Conclusions:

  • Human eosinophils express functional CCR7.
  • CCR7 mediates eosinophil responses to CCL19 and CCL21, including migration and intracellular signaling.
  • This study elucidates a novel mechanism for eosinophil trafficking in inflammatory conditions.
