Summary

We present a method for isolating and culturing mouse primary hepatocytes. These cells are viable, transfectable, and retain essential liver functions, offering a better model than liver-derived cell lines.

Area of Science:

  • Hepatology
  • Cell Biology
  • Pharmacology

Background:

  • Primary hepatocytes better model liver function in vivo compared to established liver cell lines.
  • Accurate in vitro models are crucial for studying liver physiology and drug metabolism.

Purpose of the Study:

  • To describe a reliable method for the isolation and culture of mouse primary hepatocytes.
  • To demonstrate the viability and key functional properties of these primary cells.

Main Methods:

  • Isolation of primary hepatocytes from mouse liver tissue.
  • Establishment of cell culture conditions.
  • Assessment of cell viability and DNA transfectability.
  • Evaluation of drug-inducible gene expression (cytochrome P450).

Main Results:

  • Successfully isolated and cultured mouse primary hepatocytes.
  • Demonstrated high cell viability and successful DNA transfection.
  • Confirmed retention of key liver-specific functions, including phenobarbital-induced cytochrome P450 gene expression.

Conclusions:

  • The described method yields viable and functional mouse primary hepatocytes.
  • These primary hepatocytes serve as a superior in vitro model for liver research compared to cell lines.
  • The model is suitable for studying drug metabolism and gene induction pathways in the liver.
