Ternary solid dispersions of meloxicam with moringa and polyvinylpyrrolidone (PVP) significantly enhance drug dissolution. Ball-milled meloxicam-moringa-PVP systems showed the greatest improvement in dissolution rate.
Area of Science:
- Pharmaceutical Sciences
- Materials Science
- Drug Delivery
Background:
- Poorly water-soluble nonsteroidal anti-inflammatory drugs (NSAIDs) like meloxicam often exhibit limited bioavailability.
- Solid dispersions are a promising strategy to enhance the dissolution and absorption of such drugs.
- Moringa seeds and polyvinylpyrrolidone (PVP) are explored as potential excipients for solid dispersions.
Purpose of the Study:
- To investigate the effect of ternary solid dispersions of meloxicam with moringa coagulant and PVP on in vitro dissolution properties.
- To compare the dissolution enhancement of binary and ternary systems.
- To identify the optimal formulation and preparation method for improved meloxicam dissolution.
Main Methods:
- Preparation of binary (meloxicam-moringa, meloxicam-PVP) and ternary (meloxicam-moringa-PVP) solid dispersions using physical kneading and ball milling.
- Characterization of solid-state properties using Fourier transform infrared spectroscopy, differential scanning calorimetry, and X-ray diffractometry.
- Evaluation of in vitro dissolution behavior using a United States Pharmacopeia type II dissolution apparatus.
Main Results:
- Solid-state studies confirmed strong interactions and amorphous nature in meloxicam-moringa-PVP systems.
- All ternary solid dispersions significantly improved meloxicam dissolution compared to binary systems.
- The meloxicam-moringa-PVP (1:3:1) ternary system prepared by ball milling exhibited the highest dissolution rate, approximately a six-fold increase.
Conclusions:
- Ternary solid dispersions of meloxicam with moringa and PVP are highly effective in enhancing dissolution.
- Ball milling is a superior preparation method for achieving enhanced dissolution.
- The synergistic interaction between moringa and PVP, along with the hydrophilic nature imparted by PVP, contributes to the improved dissolution of meloxicam.
