New nitrogen-containing apigenin analogs show promising anticancer, antibacterial, and antioxidant activities. Compound 4i demonstrated potent antiproliferative and antioxidant effects, while analogs 4h and 4j were effective antibacterial agents.
Area of Science:
- Medicinal Chemistry
- Organic Synthesis
- Pharmacology
Background:
- Flavonoids, like apigenin, are plant-derived compounds with potential therapeutic applications.
- Developing novel analogs of natural products can lead to improved pharmacological properties.
- Nitrogen-containing modifications of flavonoids are an area of interest for drug discovery.
Purpose of the Study:
- To synthesize novel nitrogen-containing apigenin analogs.
- To evaluate the in vitro anticancer, antibacterial, and antioxidant activities of these synthesized compounds.
- To identify lead compounds with enhanced biological activities compared to the parent apigenin.
Main Methods:
- Synthesis of apigenin analogs (4a-j) utilizing Mannich reactions.
- Structural elucidation of synthesized compounds using Nuclear Magnetic Resonance ((1)H-NMR, (13)C-NMR) and Electrospray Ionization Mass Spectrometry (ESI-MS).
- In vitro evaluation of antiproliferative activity against human cancer cell lines (HeLa, HepG2, A549, MCF-7), antioxidant activity (DPPH assay), and antibacterial activity against pathogenic bacteria (Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa).
Main Results:
- All synthesized apigenin analogs displayed varying degrees of antiproliferative activity against tested cancer cell lines.
- Compound 4i exhibited the most significant antiproliferative effects (IC50 values ranging from 40 to 223 μg/mL) and potent antioxidant activity (IC50 = 334.8 μg/mL).
- Compounds 4h and 4j demonstrated superior antibacterial activity against Gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) with low Minimum Inhibitory Concentrations (MIC values of 3.91 and 1.95 μg/mL, respectively).
Conclusions:
- The synthesized nitrogen-containing apigenin analogs possess significant anticancer, antibacterial, and antioxidant properties.
- Compound 4i is a promising candidate for further development as an anticancer and antioxidant agent.
- Analogs 4h and 4j show potential as novel antibacterial agents, particularly against Gram-positive pathogens.
