Aliasing01:18

Aliasing

Accurate signal sampling and reconstruction are crucial in various signal-processing applications. A time-domain signal's spectrum can be revealed using its Fourier transform. When this signal is sampled at a specific frequency, it results in multiple scaled replicas of the original spectrum in the frequency domain. The spacing of these replicas is determined by the sampling frequency.
If the sampling frequency is below the Nyquist rate, these replicas overlap, preventing the original signal...

Electron Microscope Tomography and Single-particle Reconstruction

Transmission electron microscopy (TEM) can be used to determine the 3D structure of biological samples with the help of techniques such as electron microscope tomography and single-particle reconstruction. While single-particle reconstruction can examine macromolecules and macromolecular complexes in vitro conditions only, tomography permits the study of cell components or small cells in vivo.
Electron Tomography
Electron tomography can be performed either in TEM or STEM (scanning transmission...

Area of Science:

  • Medical Imaging
  • Nuclear Medicine
  • Biomedical Engineering

Background:

  • Respiratory motion during SPECT/PET imaging blurs reconstructions, impairing quantitative accuracy, especially in abdominal regions.
  • Phase-based gated reconstruction methods can reduce motion artifacts but often degrade signal-to-noise ratio (SNR) and image quality.

Purpose of the Study:

  • To develop a 3D reconstruction method for micro-SPECT imaging in mice that corrects for respiratory motion without compromising image quality.
  • To improve the accuracy of quantitative measurements in micro-SPECT studies affected by respiratory motion.

Main Methods:

  • A novel 3D reconstruction approach for 4D micro-SPECT data acquisition.
  • Identification and exclusion of list-mode data associated with respiratory motion phases.
  • Reconstruction of motion-free 3D images from selected data.

Main Results:

  • The proposed method successfully reconstructs motion-free 3D micro-SPECT images.
  • Compared to standard 3D reconstruction, an increase of 14.6% in mean standardized uptake value was observed.
  • Compared to gated 4D reconstruction, the method demonstrated up to 60% less noise and a 124% increase in SNR.

Conclusions:

  • The developed 3D reconstruction method effectively eliminates respiratory motion artifacts in mouse micro-SPECT imaging.
  • This technique provides motion-free images with improved count statistics and quantitative accuracy.
  • The method offers significant advantages in noise reduction and SNR enhancement over existing gated reconstruction techniques.
