Endothelium-dependent vasodilation is impaired in spontaneously hypertensive rats (SHR), contributing to their high blood pressure. This study investigated vasodilation and hypotension in SHR versus normotensive rats.

Area of Science:

  • Cardiovascular Physiology
  • Hypertension Research

Background:

  • Spontaneously hypertensive rats (SHR) exhibit elevated blood pressure compared to normotensive Wistar-Kyoto rats (WKY).
  • Endothelial dysfunction is implicated in the pathophysiology of hypertension.

Purpose of the Study:

  • To comparatively investigate endothelium-dependent and independent hypotension, vasodilation, and relaxation in SHR and WKY rats, both in vivo and in vitro.
  • To determine the role of endothelial function in maintaining hypertension in SHR.

Main Methods:

  • In vivo studies in conscious and anesthetized rats to assess hypotension and blood flow responses to acetylcholine, adenosine triphosphate (ATP), and sodium nitroprusside.
  • In vitro studies using isolated mesenteric arteries and aorta to evaluate relaxation responses to the same agents after endothelium removal.

Related Experiment Videos

Main Results:

  • Hypotensive responses to acetylcholine and ATP were attenuated in SHR compared to WKY rats.
  • Sodium nitroprusside induced similar hypotension in both rat strains.
  • Endothelium-dependent relaxation to acetylcholine was impaired in SHR, while nitroprusside-induced relaxation was unaffected.
  • ATP-induced renal blood flow reduction was greater in SHR.

Conclusions:

  • Suppressed endothelium-dependent relaxation of resistant arterioles is linked to the maintenance of hypertension in SHR.
  • Endothelial dysfunction plays a significant role in the elevated blood pressure observed in spontaneously hypertensive rats.
