Summary

Viral encephalitis alters central serotonin function, causing presynaptic deficits and postsynaptic hyperreactivity. This study investigated these changes in mice infected with Venezuelan equine encephalitis virus.

Area of Science:

  • Neuroscience
  • Virology
  • Biochemistry

Background:

  • Viral encephalitis can impact neurological function.
  • Serotonergic system plays a crucial role in brain function and behavior.
  • Understanding neurochemical changes during viral infections is vital.

Purpose of the Study:

  • To evaluate central serotonergic function during viral encephalitis.
  • To investigate the effects of Venezuelan equine encephalitis virus (Pixuna strain) on serotonin levels and turnover.
  • To correlate neurochemical changes with behavioral and immunohistofluorescence findings.

Main Methods:

  • Biochemical assays to measure serotonin and 5-hydroxyindoleacetic acid.
  • Pharmacological manipulation using agents like pargyline, p-chlorophenylalanine, and probenecid.
  • Behavioral tests including the swim test.
  • Immunohistofluorescence to detect viral antigen distribution.

Main Results:

  • No significant changes in serotonin and 5-hydroxyindoleacetic acid levels were observed.
  • A differential decrease in serotonin turnover rate was noted in the raphe and cortex.
  • Sick animals showed a decreased ratio of serotonin turnover rate to steady-state concentration in the raphe.
  • Infected animals exhibited increased response to 5-methoxy-N,N-dimethyltryptamine and reduced immobility in the swim test.
  • Viral antigens were predominantly localized in the raphe and periraphe areas.

Conclusions:

  • The findings suggest a presynaptic serotonin deficit and postsynaptic hyperreactivity in the serotonergic system during viral encephalitis.
  • The virus distribution is region-selective, with a higher concentration in raphe and periraphe areas.
  • These neurochemical and viral distribution patterns contribute to the observed behavioral changes.
