Background:

  • The molecular basis of cancer of unknown primary (CUP) is poorly understood.

Purpose of the Study:

  • To identify a common diagnostic biomarker for cancer of unknown primary (CUP).
  • To analyze gene expression profiles in CUP tumors.
  • To explore the molecular characteristics of CUP.

Main Methods:

  • Microarray gene expression analysis of tumor mRNA from 60 CUP patients.
  • Normalization using asinh transformation to create CUP-specific gene expression profiles.
  • Comparison with non-CUP groups using t-tests to identify significant genes (p<0.001).

Main Results:

  • Identified 44 up-regulated genes in CUP, including 6 ribosomal protein genes.
  • Two ribosomal protein genes (RPS7 and RPL11) are linked to the Mdm2-p53 pathway.
  • Genes associated with metastasis and apoptosis were also identified, suggesting biological attributes of CUP.

Conclusions:

  • The identified up-regulated and down-regulated genes may serve as unique biomarkers for CUP.
  • Protein products of these genes could have clinical utility in diagnosing CUP.
  • This research provides a foundation for further investigation into CUP's molecular underpinnings.
