Area of Science:

  • Biophysics
  • Molecular Biology
  • Biochemistry

Background:

  • Single molecule photobleaching quantifies protein complex stoichiometry by counting fluorophore steps.
  • Unobserved bleaching events create binomial distributions, complicating accurate stoichiometry determination.
  • Current methods lack robust confidence quantification for interpreting photobleaching data.

Purpose of the Study:

  • To address the ill-posed inference problem in single molecule photobleaching data.
  • To develop a general inference model for interpreting photobleaching data.
  • To provide methods for accurate parameter confidence estimation in stoichiometry studies.

Main Methods:

  • Development of a general statistical inference model for binomial distributions.
