Cancer-Critical Genes II: Tumor Suppressor Genes

Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype.
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.

Summary

CD10 is expressed in seminomas and their precursors, intratubular germ cell neoplasia. This marker is lost upon differentiation, making CD10 valuable for diagnosing testicular germ cell tumors.

  • It is present in normal tissues and various cancers.
  • Its role in testicular germ cell tumors is not well understood.

Purpose of the Study:

  • Investigate CD10 expression in testicular germ cell tumors.
  • Assess the diagnostic and biologic significance of CD10.
  • Determine CD10's utility in differentiating tumor types.

Main Methods:

  • Studied 135 testicular germ cell tumor cases.
  • Utilized immunohistochemistry for CD10 detection.
  • Analyzed pure and mixed germ cell tumor forms.

Main Results:

  • CD10 was positive in 92% of seminomas.
  • 67% of the seminomatous component in mixed tumors expressed CD10.
  • Intratubular germ cell neoplasia consistently expressed CD10.
  • Anaplastic seminomas, embryonal carcinomas, teratomas, and spermatocytic seminomas were negative.

Conclusions:

  • CD10 expression is specific to seminomas and their precursors.
  • Loss of CD10 correlates with differentiation into embryonal, somatic, or trophoblastic lines.
  • CD10 is a potential marker for distinguishing seminomas and assessing mixed tumors.
