    Area of Science:

    • Immunology
    • Cell Biology

    Background:

    • Phagocytosis is a critical cellular process for immune defense.
    • Accurate quantification of phagocytic activity is essential for immunological studies.

    Purpose of the Study:

    • To develop a rapid, quantitative assay for phagocytosis.
    • To assess the phagocytic capability of human and dog polymorphonuclear leukocytes (PMN's) without serum.

    Main Methods:

    • Utilized 3H-labeled Staphylococcus albus as target particles.
    • Incubated cells at 37°C for 30 minutes in a defined medium.
    • Measured phagocyte radioactivity to quantify engulfment.

    Main Results:

    • The assay demonstrated rapid engulfment, nearing completion within 30 minutes.
    • Phagocyte radioactivity increased proportionally with polymorphonuclear leukocyte concentration.
    • Maximal phagocytic activity was observed at pH 7.5.

    Conclusions:

    • A novel, rapid, and quantitative assay for phagocytosis has been established.
    • The assay effectively measures PMN phagocytic capability in a serum-free defined medium.
    • Optimal conditions for maximal phagocytosis by PMNs were identified at pH 7.5.
