Background:

Understanding the brain mechanisms of olfaction requires deciphering the relationship between odorant molecular structure and perceived smell.
Natural odors are complex mixtures, and predicting the smell of these mixtures from their molecular composition remains a significant challenge.

Purpose of the Study:

To develop and validate a predictive model linking the molecular structure of odorant mixtures to their perceived similarity.
To investigate whether the brain processes olfactory mixtures analytically or synthetically.

Main Methods:

139 subjects rated the perceptual similarity of 64 odorant mixtures (4-43 components).
Evaluated models predicting mixture similarity based on molecular structure, comparing analytical (component-wise) and synthetic (single vector) approaches.

Main Results:

A model treating mixtures as single structural vectors significantly predicted perceptual similarity (r≥0.49, p<0.001).
An optimized single-vector model achieved high correlation (r=0.85, p<0.001) between predicted and actual mixture similarity.
The findings indicate that an algorithm can predict the perceived similarity of novel odorant mixtures based on their molecular structure.

Conclusions:

The study presents a successful algorithm for predicting olfactory mixture perception from molecular structure.
The efficacy of the single-vector model supports a synthetic processing mechanism in the brain's olfactory system, rather than an analytical one.
