Summary
Insulin receptor (IR) gene expression increases with muscle differentiation and is directly regulated by glucocorticoids. Insulin, however, decreases IR gene expression and accelerates receptor degradation in target cells.
Area of Science:
- Molecular Endocrinology
- Cell Biology
- Gene Regulation
Background:
- Insulin receptor (IR) is crucial for insulin signaling and cell function.
- Understanding IR gene regulation is key to studying insulin action and related diseases.
- Muscle and pancreatic cells are significant targets for insulin.
Purpose of the Study:
- To investigate the regulation of the insulin receptor (IR) gene during cell differentiation.
- To determine the effects of hormones, specifically glucocorticoids and insulin, on IR gene expression.
- To elucidate the mechanisms controlling IR gene transcription and expression.
Main Methods:
- Utilized BC3H1 mouse muscle cells and AR42J pancreatic acinar cells as model systems.
- Employed Northern and slot-blot analyses to study IR mRNA levels.
- Measured IR transcription using elongation assays and analyzed IR mRNA half-life.
- Investigated IR promoter activity via gene transfection with reporter plasmids (e.g., CAT assay).
Main Results:
- Cell differentiation, particularly in muscle and pancreatic cells, led to a significant increase (5-10 fold) in IR mRNA and content.
- Glucocorticoids directly stimulated IR gene transcription (5-10 fold) and increased IR mRNA levels in lymphocytes.
- Insulin accelerated IR degradation and decreased IR biosynthesis by approximately 50% in pancreatic cells, leading to reduced IR mRNA levels.
- IR mRNA was found to have multiple transcription start sites.
Conclusions:
- IR gene expression is selectively upregulated during muscle and pancreatic cell differentiation.
- Glucocorticoids directly enhance IR gene transcription, demonstrating hormonal regulation.
- Insulin downregulates IR expression by decreasing biosynthesis and accelerating degradation, indicating complex feedback mechanisms.
- The study elucidated key aspects of IR gene regulation by differentiation and hormonal factors.
