Vaccine Production01:23

Vaccine Production

158
Vaccine production involves a sequence of upstream and downstream processes to generate a safe and effective immunological product. It begins with cultivating microorganisms, such as viruses or bacteria, to obtain antigenic material. For viral vaccines, mammalian host cells are grown in bioreactors and subsequently infected with the target virus. The virus replicates within the host cells, which are lysed to release viral particles. This lysate is then clarified through filtration or...

Summary

A novel canine influenza vaccine using Malva mosaic virus (MaMV) nanoparticles and an OmpC adjuvant shows promise. This M2e peptide-based vaccine formulation is immunogenic in dogs and offers broad protection against H3N8 and other influenza strains.

Area of Science:

  • Veterinary Virology
  • Immunology
  • Vaccine Development

Background:

  • Canine H3N8 influenza emerged in 2004, necessitating new control strategies.
  • Existing vaccines may be challenged by viral mutation (drift and shift).
  • A need exists for a broadly protective vaccine against evolving canine influenza strains.

Purpose of the Study:

  • To develop a novel vaccine platform for canine influenza.
  • To enhance immunogenicity and stability of the M2e peptide antigen.
  • To achieve broad protection against diverse influenza strains in dogs.

Main Methods:

  • Utilized Malva mosaic virus (MaMV) nanoparticles as a vaccine platform.
  • Incorporated the universal M2e peptide antigen.
  • Included OmpC adjuvant purified from Salmonella typhi.
  • Evaluated vaccine immunogenicity and efficacy in mouse and dog models.

Main Results:

  • MaMV nanoparticles improved M2e peptide stability and immunogenicity.
  • OmpC adjuvant significantly boosted immune response and broadened protection.
  • Vaccine formulation demonstrated cross-reactivity with H9N2, H5N1, and H1N1 M2e peptides.
  • Optimal formulation was immunogenic in dogs.

Conclusions:

  • The MaMV-based vaccine platform effectively elicits an immune response to the M2e peptide.
  • The OmpC adjuvant enhances protection against heterosubtypic influenza strains.
  • The developed vaccine formulation shows potential for commercialization in canine influenza.
  • Broadly protective M2e-based vaccines are crucial for managing canine influenza.
