Summary

The mechanical method is superior for isolating mare ovarian preantral follicles (PAFs) compared to mechanical-enzymatic methods. Both saline solution (SS) and PBS are suitable media for preparing equine PAFs.

Background:

Purpose of the Study:

  • To evaluate mechanical and mechanical-enzymatic methods for mare PAF isolation.
  • To compare saline solution (SS) and PBS as media for PAF preparation.

Main Methods:

  • Ovarian tissues were processed using mechanical (mixer) or mechanical-enzymatic (collagenase) methods.
  • Incubation times of 10 and 20 minutes were tested.
  • Follicle counts were performed using SS and PBS solutions.

Main Results:

  • The mechanical method yielded significantly more PAFs than the mechanical-enzymatic method in both incubation groups.
  • No significant difference was observed between SS and PBS solutions for PAF isolation.
  • Mechanical isolation achieved higher follicle recovery rates.
