Ribosome Profiling02:24

Ribosome Profiling

3.2K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

Area of Science:

  • Epigenetics
  • Bioinformatics
  • Genomics

Background:

  • Reduced representation bisulfite sequencing (RRBS) generates large datasets for DNA methylation analysis.
  • Analyzing RRBS data presents significant bioinformatics challenges for researchers.
  • Understanding DNA methylation patterns is crucial for epigenetic studies.

Purpose of the Study:

  • To develop a comprehensive bioinformatics server, RRBS-Analyser, for genome-scale DNA methylation analysis using RRBS data.
  • To provide tools for assessing sequencing quality, aligning reads, and identifying/annotating methylcytosines (5mCs).
  • To enable detection, annotation, and visualization of differentially methylated regions (DMRs) across multiple samples and reference organisms.

Main Methods:

  • Development of the RRBS-Analyser server.
  • Utilizing RRBS data for DNA methylation analysis.
  • Implementation of algorithms for read alignment, 5mC identification, annotation, and DMR detection.

Main Results:

  • RRBS-Analyser assesses sequencing quality and provides statistical information.
  • The server identifies and annotates 5mCs in CG, CHG, and CHH contexts, linking them to genomic features.
  • It supports detection, annotation, and visualization of DMRs and provides gene annotations for DMR-containing genes.

Conclusions:

  • RRBS-Analyser offers a comprehensive solution for analyzing RRBS data, addressing bioinformatics challenges in DNA methylation studies.
  • The tool facilitates the identification and annotation of methylcytosines and differentially methylated regions.
  • RRBS-Analyser aids epigenetic researchers in unraveling DNA methylome complexities and supports downstream gene-specific studies.
