Labeling DNA Probes03:31

Labeling DNA Probes

7.8K
DNA probes are fragments of DNA labeled with a reporter tag to enable their detection or purification. The resulting labeled DNA probes can then hybridize to target nucleic acid sequences through complementary base-pairing, and may be used to recover or identify these regions.
Radioisotopes, fluorophores, or small molecule binding partners like biotin or digoxigenin, are the most widely used reporter tags for labeling DNA probes. These labels can be attached to the probe DNA molecule via...

Area of Science:

  • Microbiology
  • Molecular Biology
  • Biotechnology

Context:

  • Accurate detection of Streptococcus mutans is crucial for managing dental caries.
  • Conventional PCR methods can suffer from cross-reactivity, leading to false positives.
  • Developing highly specific diagnostic tools is essential in clinical microbiology.

Purpose:

  • To design and validate a novel TaqMan® MGB probe for specific Streptococcus mutans identification.
  • To compare the specificity and sensitivity of the new TaqMan® MGB real-time PCR assay against conventional nested PCR.
  • To establish a reliable molecular method for detecting Streptococcus mutans in clinical samples.

Summary:

  • A new TaqMan® MGB probe was designed based on sequence analysis of the 16S rRNA gene.
  • Conventional nested PCR showed cross-reactivity with Streptococcus gordonii, yielding false positives.
  • The developed TaqMan® MGB real-time PCR assay exclusively detected Streptococcus mutans, demonstrating high specificity.
  • The detection limit for Streptococcus mutans using TaqMan® MGB real-time PCR was determined to be 20 μg/L.

Impact:

  • The novel TaqMan® MGB probe significantly improves the specificity of Streptococcus mutans detection.
  • This validated real-time PCR method offers a sensitive and accurate alternative for identifying Streptococcus mutans.
  • The findings contribute to the development of more precise diagnostic tools for oral bacteria and related diseases.
