    Background:

    • Cryosurgery is a cancer treatment that uses extreme cold to destroy abnormal cells.
    • Understanding the vascular response to cryosurgery is crucial for optimizing combination therapies.

    Purpose of the Study:

    • To investigate the combined effects of cryosurgery and anticancer drugs (cryochemotherapy) on a B16 melanoma tumor model.
    • To assess the vascular changes in normal tissue and tumors following cryosurgery.
    • To determine the optimal timing for administering anticancer drugs in conjunction with cryosurgery.

    Main Methods:

    • Cryosurgery was performed on a B16 melanoma/BDF1 tumor system.
    • Vascular volume and permeability were measured using radiolabeled red blood cells (51Cr) and serum albumin (125I).
    • Anticancer drugs (peplomycin, adriamycin) were administered at various times relative to cryosurgery, and drug concentrations in tumors were analyzed.

    • Cryosurgery initially increased vascular volume and permeability in both normal tissue and tumors, with subsequent decreases.

    • Administering peplomycin 5 minutes to 3 hours after cryosurgery resulted in higher tumor drug concentrations.
    • Peplomycin administered before cryosurgery and adriamycin at any time did not show significant tumor trapping.

    Conclusions:

    • There are distinct functional differences in vascular response to cryoinjury between normal tissue and tumors.
    • The timing of anticancer drug administration is critical for effective cryochemotherapy.
    • Cryochemotherapy, by enabling sustained high drug concentrations in tumors, presents a novel approach for multidisciplinary cancer treatment.
