DNA Isolation01:24

DNA Isolation

35.4K
DNA isolation protocols can be fast and straightforward or complex and time-consuming depending on the type and quality of DNA required for further processing. For example, plasmid DNA extraction is a bit more complicated than genomic DNA extraction because of the need for an appropriate lysis method to separate plasmid DNA from gDNA during isolation. However, for specific applications, such as long-range DNA sequencing that require a good yield of high- quality DNA samples, we need to follow...

DNA Isolation

177.4K
DNA from cells is required for many biotechnology and research applications, such as molecular cloning. To remove and purify DNA from cells, researchers use various methods of DNA extraction. While the specifics of different protocols may vary, some general concepts underlie the process of DNA extraction.

Summary

Researchers successfully extracted DNA from stored urine samples of patients with ANCA-associated vasculitis. This method provides a valuable alternative for DNA analysis when other samples are unavailable, aiding in database completion.

Area of Science:

  • Nephrology
  • Molecular Biology
  • Genetics

Background:

  • Traditionally, DNA for analysis is obtained from buffy coats.
  • DNA in urine is often disregarded due to perceived rapid degradation.
  • This study aimed to retrieve DNA from stored urine for patient databases.

Purpose of the Study:

  • To assess the feasibility of DNA extraction from stored urine samples.
  • To establish a method for DNA recovery from urine for ANCA-associated vasculitis patient databases.

Main Methods:

  • Urine samples were collected from patients with informed consent.
  • Samples underwent filtration, dialysis, concentration, and freeze-drying.
  • DNA extraction utilized the high pure PCR template preparation kit, followed by purity and concentration analysis.

Main Results:

  • DNA was successfully extracted from 86.8% of processed stored urine samples.
  • The extracted DNA had a mean concentration of 258.7 ng/μL and a purity ratio of 1.81.
  • This DNA was confirmed to be usable for PCR analysis.

Conclusions:

  • DNA can be retrieved from room-temperature stored urine using a specific processing method.
  • This technique enables DNA recovery even after prolonged storage, contrary to previous assumptions.
  • This method is a viable option for genetic analysis when other biological samples are inaccessible.
