Background:

  • Chromosome 1R in rye (Secale cereale) is important for agriculture.
  • Previous genetic maps of rye chromosome 1R have limitations in resolution and marker order.

Purpose of the Study:

  • To construct a high-resolution genetic map of rye chromosome 1R.
  • To order known-function loci and anonymous clones along chromosome 1R.
  • To compare the newly constructed map with existing rye chromosome maps.

Main Methods:

  • Utilized 16 molecular and biochemical loci for mapping.
  • Integrated known-function loci and anonymous genomic and cDNA clones from wheat.
  • Calculated map distances in centimorgans (cM).

Main Results:

  • A genetic map spanning 106 cM was constructed for rye chromosome 1R.
  • Twelve loci were found to be clustered within a 15-cM region around the centromere.
  • The order of loci from the long arm to the short arm was established: XAdh - XLee - Glu-R1[Sec-3] - XPpdk-1R - XEm-1R-1 - XEm-1R-2 - Centromere - XNor-R1 -Gpi-R1 - XGli-R1 [Sec-1a].
  • The centromeric region showed good agreement with previous maps.
  • A significantly larger genetic distance was observed for the XNor-R1 - Gpi-R1 region compared to prior reports.

Conclusions:

  • The new map provides an improved framework for rye chromosome 1R genetics.
  • Discrepancies in the XNor-R1 - Gpi-R1 region suggest potential for further refinement of rye genetic maps.
  • This map can facilitate comparative mapping studies between rye and other Triticeae species.
