Summary

Background:

Posttraumatic stress disorder (PTSD) development involves environmental and genetic factors.
The 5-HTTLPR polymorphism in the serotonin transporter gene is a candidate genetic risk factor for PTSD.

Purpose of the Study:

To investigate the association between 5-HTTLPR gene variation and PTSD risk.
To resolve controversial findings and increase statistical power through meta-analysis.

Main Methods:

A meta-analysis of 12 studies involving trauma-exposed samples published up to December 2012.
Searched PubMed and ISI databases; assessed study quality and publication bias.
Used Cochrane Collaboration Review Manager and Comprehensive Meta-Analysis software for data analysis and metaregression.

Main Results:

No overall association was found between 5-HTTLPR and PTSD (p = .10), with significant heterogeneity.
Sensitivity analyses revealed an association between the SS genotype and PTSD in participants with high trauma exposure (p < .001).
Gender differences may partly explain the observed heterogeneity.

Conclusions:

Carrying the SS genotype appears to be a risk factor for PTSD specifically in individuals with high trauma exposure.
Further research into Gene × Environment interactions is crucial for understanding this polymorphism's role in PTSD.
