In-situ Hybridization02:31

In-situ Hybridization

9.2K
In situ hybridization (ISH) is a technique used to detect and localize specific DNA or RNA molecules in cells, tissue, or tissue sections using a labeled probe. The technique was first used in 1969 for the investigation of nucleic acids. It is currently an essential tool in scientific research and clinical settings, especially for diagnostic purposes.
Types of probes and labels
A probe is a complementary strand of DNA or RNA that binds to corresponding nucleotide sequences in a cell. Many...

Magnetic bacteria exhibit a directed movement called magnetotaxis, driven by structures called magnetosomes. These magnetosomes consist of chains of magnetic particles made of either magnetite (Fe₃O₄) or greigite (Fe₃S₄) and are organized in a linear conformation by a protein scaffold within invaginations of the cell membrane. The bacteria align along the north–south magnetic field lines, much like a compass needle. They are typically microaerophilic or anaerobic...

Diversity of Protists IV01:27

Diversity of Protists IV

2.1K
Amoebozoa represent a diverse group of terrestrial and aquatic protists that utilize lobe-shaped pseudopodia for locomotion and feeding. This characteristic differentiates them from the Rhizaria, which possess threadlike pseudopodia. The primary classifications within Amoebozoa include gymnamoebas, entamoebas, and the plasmodial and cellular slime molds. Phylogenetic evidence indicates that Amoebozoa diverged from a lineage that ultimately gave rise to fungi and animals.Gymnamoebas and...

Summary

A novel actinobacterial strain, Kocuria indica sp. nov., was discovered in Indian sediment. This Gram-positive coccoid bacterium is phylogenetically distinct from other Kocuria species based on 16S rRNA gene analysis and DNA-DNA hybridization.

Area of Science:

  • Microbiology
  • Bacteriology
  • Taxonomy

Background:

  • Actinobacteria are a diverse phylum of bacteria with significant ecological and biotechnological roles.
  • The genus Kocuria comprises Gram-positive, aerobic cocci, with several species isolated from various environments.
  • Accurate taxonomic classification is crucial for understanding microbial diversity and function.

Purpose of the Study:

  • To isolate and taxonomically characterize a novel actinobacterial strain from a sediment sample.
  • To determine the phylogenetic and chemotaxonomic properties of the isolate.
  • To propose a new species within the genus Kocuria.

Main Methods:

  • Isolation of actinobacteria from sediment samples.
  • Polyphasic taxonomic study including phenotypic, chemotaxonomic, and molecular analyses.
  • 16S rRNA gene sequencing and DNA-DNA hybridization for phylogenetic placement.
  • Analysis of cellular fatty acids, phospholipids, and menaquinones.

Main Results:

  • A novel Gram-positive, aerobic, non-motile coccoid strain, NIO-1021(T), was isolated.
  • Phylogenetic analysis based on 16S rRNA gene sequences showed 98.6% similarity to Kocuria marina.
  • DNA-DNA hybridization and chemotaxonomic data supported the differentiation from related Kocuria species.
  • The DNA G+C content was 60.5 mol%.

Conclusions:

  • Strain NIO-1021(T) represents a novel species of the genus Kocuria.
  • The proposed name for the novel species is Kocuria indica sp. nov.
  • Kocuria indica sp. nov. (type strain NIO-1021(T)) expands the known diversity within the Kocuria genus.
