Do columnar cell lesions exist in the male breast?

Columnar cell lesions (CCLs) are linked to female breast cancer, but this study found no convincing CCLs in male breast cancer, gynaecomastia, or normal male breast tissue, suggesting they are uncommon in male breast carcinogenesis.

- Immunohistochemistry for cytokeratin 5/6 (CK5/6), CK14, and oestrogen receptor alpha (ER) was performed.
- Analysis focused on identifying CCL morphology and ductal epithelium characteristics.
Main Results:
- No lesions with convincing CCL morphology were identified in male breast cancer, gynaecomastia, or normal male breast tissue.
- Some ducts in male breast cancer showed features suggestive of CCLs but lacked typical morphology.
- Three-layered ductal epithelium, characteristic of gynaecomastia, was frequently observed.
- A few ducts in male breast cancer were found to be clonally negative for basal cytokeratins.
Conclusions:
- CCLs appear to be very uncommon in the male breast.
- These lesions are unlikely to play a significant role in male breast carcinogenesis.
- Further research may be needed to definitively exclude the existence of CCLs in males.
