Mouse beta-interferon reduces RNA efflux from isolated nuclei.

H S MacDonald, V M Kushnaryov, R Hevner

    Journal of Interferon Research
    |June 1, 1986

    Summary

Mouse interferon-beta (MuIFN-beta) was found to directly impact the cell nucleus. This antiviral protein reduced the release of RNA from L929 cell nuclei in a dose-dependent manner.

    Area of Science:

    • Cell Biology
    • Virology
    • Molecular Biology

    Background:

    • Interferons (IFNs) are critical antiviral proteins.
    • The precise nuclear mechanisms of IFN action are not fully understood.

    Purpose of the Study:

    • To investigate the direct effects of MuIFN-beta on isolated L929 cell nuclei.
    • To determine if MuIFN-beta influences RNA efflux from the nucleus.

    Main Methods:

    • Isolated L929 cell nuclei were treated with varying doses of MuIFN-beta.
    • The energy-dependent efflux of radiolabeled RNA (3H-RNA) from the nuclei was measured.

    Main Results:

    • MuIFN-beta treatment reduced the efflux of 3H-RNA from L929 cell nuclei.
    • This reduction was observed in a dose-dependent manner, indicating a specific interaction.

    Conclusions:

    • MuIFN-beta exerts a direct physiological effect on the cell nucleus.
    • IFN-beta may regulate nuclear RNA export or stability, contributing to its antiviral activity.
