Enlargement of the Plasma Membrane01:22

Enlargement of the Plasma Membrane

1.8K
Cell division and enlargement are processes that require precise control. The control ensures that cell division cannot proceed unless the cell has grown to a specific size. A spherical, dividing cell requires an approximately 1.6X increase in its surface area to double its volume. The secretory pathway also has a significant role in cell membrane enlargement. Secretory vesicles that bud off from the Golgi apparatus and later fuse with the plasma membrane during exocytosis are a major source of...

Fluid Mosaic Model01:19

Fluid Mosaic Model

14.6K
Scientists identified the plasma membrane in the 1890s and its principal chemical components (lipids and proteins) by 1915. The model for plasma membrane structure, proposed in 1935 by Hugh Davson and James Danielli, was the first model to be widely accepted in the scientific community. The model was based on the plasma membrane's "railroad track" appearance in early electron micrographs. Davson and Danielli theorized that the plasma membrane's structure resembled a sandwich...

What are Membranes?01:24

What are Membranes?

14.5K
A cell's plasma membrane demarcates the cell's borders and determines the nature of its interaction with the environment. Cells exclude certain substances, take in others, and excrete some others in controlled quantities. The plasma membrane must be flexible to allow certain cells, such as red and white blood cells, to change their shape while passing through narrow capillaries. These are the more obvious plasma membrane functions. In addition, the plasma membrane's surface carries...

The Fluid Mosaic Model01:34

The Fluid Mosaic Model

157.4K
The fluid mosaic model was first proposed as a visual representation of research observations. The model comprises the composition and dynamics of membranes and serves as a foundation for future membrane-related studies. The model depicts the structure of the plasma membrane with a variety of components, which include phospholipids, proteins, and carbohydrates. These integral molecules are loosely bound, defining the cell’s border and providing fluidity for optimal function.

Structure of Porins01:21

Structure of Porins

3.0K
Mitochondria, chloroplasts, and gram-negative bacteria have transmembrane, beta-barrel proteins called porins to mediate the free diffusion of ions and metabolites across the membrane. Mitochondrial porin precursors contain conserved amino acid sequences called beta signals at their C-terminal. Beta signals have a  motif of PoXGXXHyXHy (Po-Polar, X-Any amino acid, G-Glycine, Hy-LargeHydrophobic), which are crucial for precursor recognition to initiate precursor assembly. Beta-barrel...

Membrane Asymmetry Regulating Transporters01:19

Membrane Asymmetry Regulating Transporters

6.0K
Enzymes like flippase, floppase, and scramblase transfer phospholipids from one layer to another in the membrane, thereby affecting membrane asymmetry.
Flippase
Eukaryotic flippases are type-IV P-type ATPases or P4-ATPases belonging to P-type ATPase family proteins that are membrane-bound pumps involved in the ATP-mediated transport of ions and molecules across the membrane. Flippases flip specific phospholipids from the outer to the inner leaflet of a membrane. All P4-ATPases have one...

Area of Science:

  • Plant biochemistry
  • Cell biology
  • Membrane protein analysis

Background:

  • NADP-dependent malic enzyme (malate dehydrogenase) is a key enzyme in plant metabolism.
  • Its localization within plant cells is not fully understood, particularly its association with cellular membranes.

Purpose of the Study:

  • To identify and characterize the cellular location of NADP-dependent malic enzyme activity in plant tissues.
  • To investigate the properties of the enzyme when associated with cellular membranes.

Main Methods:

  • Isolation and characterization of membrane fractions from plant tissues (Cucurbita hypocotyls, maize coleoptiles).
  • Enzyme assays for NADP-dependent malic enzyme activity.
  • Sucrose gradient centrifugation and analysis of enzyme markers.

  • Assessment of enzyme binding stability and release under various conditions (salts, EDTA, Triton-X-100, hypotonic treatment).
  • Main Results:

    • A distinct membrane fraction containing significant NADP-dependent malic enzyme activity (up to 10% of total activity) was isolated.
    • This fraction, termed the "malic enzyme particle," was identified as the plasma membrane based on sedimentation, density, enzyme markers, and Metrizamide penetration.
    • The enzyme binding to the plasma membrane was stable but could be released by Triton-X-100 or hypotonic conditions, suggesting vesicle formation during homogenization.
    • Low malic enzyme activities were also found associated with proplastids.

    Conclusions:

    • NADP-dependent malic enzyme is localized to the plasma membrane in plant tissues.
    • The enzyme is stably bound to the plasma membrane, likely within osmotically sensitive vesicles formed during tissue homogenization.
    • This finding provides new insights into the subcellular distribution and potential roles of malic enzyme in plant cells.
