β-adrenoceptors have varied sensitivities towards adrenaline, noradrenaline, and isoprenaline. The order of agonist potency is as follows:
Isoprenaline > Adrenaline > Noradrenaline
Neurotransmitter binding to these receptors causes activation of adenylyl cyclase resulting in increased concentrations of cAMP and modulation of calcium ion channels within the cell. They are further classified into β1, β2, and β3 subtypes.

Summary

Brown adipose tissue (BAT) surrounding pheochromocytomas (PHEO) expresses key fat-burning genes. Catecholamines from PHEO influence BAT activity and adiponectin levels, which decrease after tumor removal.

Background:

  • The presence and function of human brown adipose tissue (BAT) remain subjects of investigation.
  • Pheochromocytomas (PHEO) are rare neuroendocrine tumors associated with hypertension.
  • Investigating the microenvironment of PHEO may reveal insights into adipose tissue activity.

Purpose of the Study:

  • To determine the presence and activity of BAT surrounding adrenal PHEO.
  • To analyze the expression of key genes in BAT and white adipose tissue (WAT) adjacent to PHEO.
  • To explore the relationship between catecholamines, adiponectin, and BAT markers in PHEO patients.

Main Methods:

  • Laser capture microdissection of adipose tissue (BAT and WAT) surrounding PHEO.
  • Analysis of uncoupling protein (UCP)-1 and adiponectin mRNA expression.
  • Correlation analysis of gene expression with urinary catecholamines and serum adiponectin levels before and after adrenalectomy.

Main Results:

  • BAT surrounding PHEO exhibited significantly higher adiponectin and UCP-1 mRNA expression compared to WAT.
  • Adiponectin mRNA levels correlated strongly with urinary metanephrines, vanillylmandelic acid (VMA), and serum adiponectin.
  • Serum adiponectin levels decreased significantly after adrenalectomy in PHEO patients.

Conclusions:

  • BAT adjacent to PHEO is metabolically active, expressing UCP-1 and adiponectin.
  • PHEO-derived catecholamines influence the expression of adiponectin and UCP-1 in surrounding BAT.
  • Adiponectin levels are modulated by PHEO and decrease post-surgical removal, suggesting a link between the tumor and systemic adiponectin regulation.
