    Substance P triggers postmortem bronchoconstriction in guinea pig lungs. This airway spasm is mediated by sensory nerve endings releasing substance P via an axonal reflex.

    Background:

    Purpose of the Study:

    • To investigate the role of substance P in initiating postmortem bronchoconstriction in guinea pigs.
    • To explore the involvement of neural reflexes in mediating this airway spasm.

    • Six experimental groups of guinea pigs were used, including controls and various treatments (morphine, substance P, capsaicin, tetrodotoxin, chlorisondamine).
    • Pressure-volume curves were analyzed before and after pulmonary perfusion to quantify bronchoconstriction.
    • Bronchoconstriction was indexed by the decrease in inflation volume.

    • Control animals exhibited significant postmortem bronchoconstriction.
    • Morphine attenuated the spasm, while exogenous substance P enhanced it.
    • Tetrodotoxin (TTX) significantly reduced acute capsaicin-induced bronchoconstriction, but chlorisondamine did not.

    • Substance P is the primary initiator of massive postmortem bronchoconstriction in guinea pig lungs.
    • This effect is mediated by the release of substance P from sensory nerve endings through an axonal reflex.
