    Summary

    Variant surface glycoprotein (VSG) mRNA turnover in Trypanosoma brucei is rapidly reduced during cell differentiation. This regulation, along with decreased transcription, controls VSG gene expression.

    Background:

    • Trypanosoma brucei undergoes differentiation from bloodstream to procyclic forms.
    • Variant surface glycoprotein (VSG) expression is critical for immune evasion in bloodstream forms.

    Purpose of the Study:

    • To investigate the regulation of VSG mRNA turnover during Trypanosoma brucei differentiation.
    • To compare VSG mRNA regulation with other RNA species during transformation.
    • To elucidate the molecular mechanisms controlling VSG gene expression shutoff.

    Main Methods:

    • Approach-to-equilibrium labeling and pulse-chase experiments were used.
    • In vitro transformation of bloodstream forms to procyclic cells was induced.
    • RNA stability and transcription rates of various mRNAs were analyzed.

    Main Results:

    • VSG mRNA half-life decreased significantly (from 4.5 h to 1.2 h) during transformation.
    • VSG mRNA transcription decreased ~25-fold, leading to a 100-fold reduction in steady-state levels.
    • Beta-tubulin mRNA stability increased in procyclic cells, while mini-exon-derived RNA (medRNA) remained stable.
    • Inhibition of protein synthesis also reduced VSG mRNA synthesis and stability.

    Conclusions:

    • VSG mRNA turnover is tightly regulated during Trypanosoma brucei differentiation.
    • Changes in regulatory molecules likely control both VSG gene transcription and mRNA stability.
    • Differential regulation of mRNA stability contributes to stage-specific gene expression in trypanosomes.
