    Summary

Griffonia simplicifolia (GSA-I) and soybean (SBA) lectins specifically label human eosinophils. These fluorochrome-conjugated lectins are effective diagnostic tools for identifying eosinophils in various sample types.

    Area of Science:

    • Immunology
    • Cell Biology
    • Histology

    Background:

    • Human eosinophils play a role in immune responses and allergic reactions.
    • Specific markers are needed for accurate eosinophil identification in diagnostic pathology.
    • Griffonia simplicifolia (GSA-I) and soybean (SBA) lectins are known to bind to eosinophils.

    Purpose of the Study:

    • To evaluate the utility of fluorochrome-labeled GSA-I and SBA for specific eosinophil labeling.
    • To demonstrate the application of these lectins in different biological sample preparations.

    Main Methods:

    • Preparation of fluorochrome-labeled GSA-I and SBA.
    • Application of labeled lectins to paraffin-embedded tissue sections.
    • Application of labeled lectins to peripheral blood smears.
    • Application of labeled lectins to cell suspensions for flow cytometry.

    Main Results:

    • Fluorochrome-labeled GSA-I and SBA demonstrated specific labeling of human eosinophils.
    • Effective eosinophil labeling was achieved in tissue sections, blood smears, and flow cytometry samples.
    • The lectins allowed for selective visualization of eosinophils.

    Conclusions:

    • Fluorochrome-conjugated GSA-I and SBA are reliable reagents for specific eosinophil identification.
    • These lectins can be used as diagnostic tools in histopathology and flow cytometry.
    • They facilitate further characterization of eosinophil cytoplasmic components.
