Pharmacogenomics: Identification of New Drug Targets01:29

Pharmacogenomics: Identification of New Drug Targets

121
Advances in genomics have profoundly influenced drug discovery by increasing both the speed and accuracy of pharmaceutical development. Pharmacogenomics, which examines how genetic variation influences drug response, facilitates the identification of novel therapeutic targets and enables patient stratification for personalized treatment. These strategies contribute to improved drug efficacy, minimized adverse effects, and more efficient clinical trial design.Mapping genetic differences...

Drug Discovery: Overview

10.4K
Drug discovery is a multifaceted process involving extensive screening, testing, and optimization of lead compounds to identify potential new drugs for therapeutic use. It combines several approaches, including screening large numbers of natural products, chemical modification of known active molecules, identification of new drug targets, and rational design based on biological mechanisms and drug-receptor structure. These approaches are carried out in both academic research laboratories and...

Site-Targeted Drug Delivery Systems: Polymeric Carriers

160
Polymeric carriers enhance targeted drug delivery by increasing efficacy while minimizing off-target effects. These carriers comprise a biodegradable polymeric backbone integrated with functional elements that enable targeting, improve physicochemical properties, and regulate drug release.Targeting MechanismsThe targeting ability of polymeric carriers is mediated by a homing device, which is a molecular recognition component designed to selectively bind to specific tissues or cells. Monoclonal...

CRISPR

46.4K
Genome editing technologies allow scientists to modify an organism’s DNA via the addition, removal, or rearrangement of genetic material at specific genomic locations. These types of techniques could potentially be used to cure genetic disorders such as hemophilia and sickle cell anemia. One popular and widely used DNA-editing research tool that could lead to safe and effective cures for genetic disorders is the CRISPR-Cas9 system. CRISPR-Cas9 stands for Clustered Regularly Interspaced...

CRISPR/Cas9 Genome Editing

3.2K
The CRISPR-Cas system serves as a bacterial defense mechanism against invading genetic elements such as viruses and plasmids, forming the foundation for its adaptation as a powerful genome-editing tool. Originally discovered in prokaryotes, this system has been repurposed to revolutionize genetic engineering across a wide range of organisms, including plants, animals, and humans. The core component, Cas9, is an endonuclease derived from Streptococcus pyogenes, capable of introducing...

iPS Cell Differentiation

2.2K
The ability of induced pluripotent stem cells or iPSCs to differentiate into most body cell types has stimulated repair and regenerative medicine research over the past few decades. iPSC-derived blood cells, hepatocytes, beta islet cells, cardiomyocytes, neurons, and other cell types can repair injuries or regenerate damaged tissue in diseases such as diabetes and neurodegenerative disorders.

Area of Science:

  • Biochemistry
  • Genetics
  • Pharmacology

Background:

  • Proprotein convertase subtilisin/kexin type 9 (PCSK9) plays a key role in cholesterol metabolism by promoting low-density lipoprotein receptor (LDLR) degradation.
  • Gain-of-function mutations in PCSK9 cause autosomal dominant hypercholesterolemia, while loss-of-function mutations are linked to lower LDL cholesterol and reduced coronary heart disease risk.

Purpose of the Study:

  • To evaluate PCSK9 inhibition as a therapeutic strategy for lowering LDL cholesterol.
  • To assess the efficacy and tolerability of monoclonal antibodies targeting PCSK9.

Main Methods:

  • Review of human data from phase II studies involving monoclonal antibodies (alirocumab and evolocumab) that inhibit PCSK9 binding to LDLR.
  • Evaluation of efficacy in hypercholesterolemic patients, including those with familial hypercholesterolemia, statin intolerance, and in monotherapy.

Main Results:

  • Monoclonal antibodies targeting PCSK9 demonstrated effective reduction of atherogenic lipoproteins.
  • LDL cholesterol reduction of up to 70% was observed with high-dose treatments.
  • These agents were generally well tolerated in phase II studies.

Conclusions:

  • PCSK9 inhibition represents a promising therapeutic strategy for lowering LDL cholesterol.
  • Monoclonal antibodies targeting PCSK9 are effective and well-tolerated, offering a new option for managing hypercholesterolemia.
  • Ongoing phase III trials will further elucidate the long-term efficacy and safety of these agents.
