Background:

  • Oxidative stress (OS) is a key factor in liver failure.
  • The liver's role as a primary source of OS requires investigation.

Purpose of the Study:

  • To assess OS and antioxidant status in an anhepatic porcine model.
  • To determine if the liver is the main contributor to OS.

Main Methods:

  • Total hepatectomy was performed on six pigs; five underwent sham surgery.
  • Plasma concentrations of malondialdehyde, xanthine oxidase, superoxide dismutase, and ferric reducing ability were measured.
  • Samples were collected pre-surgery and at 8 and 16 hours post-surgery.

Main Results:

  • Anhepatic pigs showed increased malondialdehyde postoperatively and at 8 hours.
  • Xanthine oxidase activity initially rose post-hepatectomy but normalized later.
  • Ferric reducing ability significantly increased in anhepatic animals, correlating positively with malondialdehyde levels.

Conclusions:

  • Hepatectomy does not eliminate oxidative stress.
  • These findings suggest significant production and regulation of OS occur outside the liver.
