Polarization changes in light diffracted from contracting muscle fibers.

Muscle contraction involves complex molecular interactions.
Understanding these dynamics requires advanced measurement techniques.

Purpose of the Study:

To investigate the dynamic changes in light polarization during muscle fiber contraction.
To correlate these optical changes with molecular events like myosin movement and calcium ion concentration.

Main Methods:

Isolated frog semitendinosus muscle fibers were used.
He-Ne laser illumination and a photoelastic modulator varied light polarization.
Diffracted light polarization was measured with 1 ms resolution during isometric contraction.
Tension, sarcomere length, and diffraction intensity were simultaneously recorded.

Main Results:

The degree of polarized light diffracted from active muscle fibers showed a biphasic variation compared to resting fibers.
Analysis linked this polarization transient to the movement of heavy meromyosin towards actin.
Increased myoplasmic calcium ion concentration was also identified as a key factor.

Conclusions:

A quantitative theory for polarized diffraction from muscle fibers was developed.
The theory accurately describes the experimental measurements.
This optical method provides a novel way to study muscle contraction dynamics.
