Genetic variations in the PERIOD3 gene influence sleep deprivation effects. PER3(5/5) carriers show greater performance decline but better sleep initiation compared to PER3(4/4) carriers.
Area of Science:
- Chronobiology
- Sleep Medicine
- Human Genetics
Background:
- Sleep deprivation significantly impairs daily functioning, with considerable individual differences in response.
- Genetic factors, such as PERIOD3 gene polymorphisms, contribute to vulnerability to sleep loss and circadian disruption.
Purpose of the Study:
- To investigate how different sleep pressure levels affect neurobehavioral performance, sleepiness, and nap sleep in individuals with varying PERIOD3 genotypes.
- To compare the impact of sleep deprivation on PER3(5/5) and PER3(4/4) allele carriers.
Main Methods:
- 14 PER3(5/5) and 15 PER3(4/4) homozygous individuals underwent 40-hour sleep deprivation and napping protocols.
- Subjective/ocular sleepiness, vigilance (psychomotor vigilance task), and unintentional sleep were assessed.
- Nap sleep was monitored using polysomnography and scored conventionally.
Main Results:
- PER3(5/5) carriers exhibited more pronounced impairments in vigilance and sleepiness correlates under high sleep pressure.
- Nap sleep efficiency and subjective sleep quality were generally higher in PER3(5/5) carriers.
- Attentional lapses under sleep deprivation correlated with sleep efficiency, suggesting a link between sleep initiation ability and vulnerability.
Conclusions:
- PER3(5/5) carriers are more susceptible to the detrimental effects of sleep deprivation on performance compared to PER3(4/4) carriers.
- Despite greater performance deficits, PER3(5/5) carriers demonstrated superior sleep initiation capabilities, possibly due to faster sleep pressure accumulation.
- Nap sleep efficiency may serve as a biomarker for individual vulnerability to elevated sleep pressure.
