Bacillus anthracis lethal toxin induces cell-type-specific cytotoxicity in human lung cell lines
1Department of Life Science, Sogang University, Shinsu-Dong, Mapo, Seoul, Korea.
Anthrax lethal toxin specifically harms human small airway epithelial cells, but not other lung cell types. This cell-specific toxicity may explain how inhalational anthrax progresses in the lungs.
Area of Science:
- Pulmonary toxicology
- Infectious disease pathology
- Cellular biology
Background:
- Inhalational anthrax, caused by Bacillus anthracis, leads to lung injury.
- Anthrax lethal toxin is known to be cytotoxic to macrophages.
- The effect of anthrax toxin on various lung cell types remains unclear.
Purpose of the Study:
- To investigate the cytotoxic effects of anthrax lethal toxin on different human lung cell types.
- To determine if anthrax toxin receptor expression correlates with cell susceptibility.
Main Methods:
- Utilized model cell lines representing lung compartments.
- Assessed cell response via MTT metabolism, neutral red uptake, and apoptosis.
- Measured anthrax toxin receptor expression and binding activity.
Main Results:
- Human small airway epithelial cells (HSAEC) were susceptible to anthrax lethal toxin.
- Other lung cell lines (A549, MRC-5, H358, SKLU-1) showed resistance.
- Anthrax toxin receptors were detected in all tested cell lines, regardless of susceptibility.
Conclusions:
- Anthrax lethal toxin exhibits cell-type-specific toxicity in human lung tissues.
- Toxicity does not correlate with anthrax toxin receptor expression levels.
- This cell-specific cytotoxicity may drive lung disease progression in inhalational anthrax.

Lethal Alleles
Lucien Cuénot discovered lethal alleles in 1905 while studying the inheritance of coat color in mice. The agouti gene is responsible for the color of the coat in mice. This gene codes for an agouti-signaling protein, which is responsible for melanin distribution in mammals. The wild-type allele gives rise to gray-brown coat color in mice, while the mutant allele gives rise to yellow coat color. In addition to coat color, the agouti gene is associated with the yellow...
