Summary

This study shows that CD8 T cells generated during tuberculosis exhibit antigen-specific killing of infected macrophages. These cells are crucial for the immune response to Mycobacterium tuberculosis.

Background:

  • Tuberculosis (TB) is a significant global health challenge caused by Mycobacterium tuberculosis.
  • Understanding the cellular immune response, particularly T cell activity, is critical for developing effective TB treatments.

Purpose of the Study:

  • To characterize the in vitro biological activities of mycobacteria-reactive Lyt-2+ T cells.
  • To investigate the cytotoxic potential and cytokine production of T cells during tuberculosis infection.

Main Methods:

  • Mice were immunized with Mycobacterium tuberculosis or M. bovis.
  • T cells were restimulated in vitro and cloned using limiting dilution.
  • Antigen-specific lysis of mycobacteria-primed macrophages and interferon-gamma production were assessed.

Main Results:

  • Established Lyt-2+ T cell lines demonstrated antigen-specific lysis of mycobacteria-primed macrophages.
  • Cytolytic activity was observed to be both MHC class I restricted and H-2 unrestricted.
  • Several T cell lines produced interferon-gamma and induced tuberculostatic macrophage activity via lymphokines or direct cell contact.

Conclusions:

  • CD8 T cells with antigen-specific cytolytic activity are generated during tuberculosis.
  • These CD8 T cells play a role in the immune response to tubercle bacilli, contributing to macrophage-mediated bacterial control.
