Certain NOD2 gene polymorphisms, specifically rs2066844, rs2066845, and rs2066847, may increase cancer risk. However, the rs2066842 polymorphism showed no significant association with cancer susceptibility in this meta-analysis.
Area of Science:
- Genetics
- Oncology
- Molecular Biology
Background:
- Emerging evidence suggests NOD2 gene polymorphisms may influence cancer susceptibility.
- Previous studies on the association between NOD2 polymorphisms and cancer risk have yielded inconclusive results.
Purpose of the Study:
- To conduct a meta-analysis to determine if NOD2 gene polymorphisms are associated with an increased risk of developing cancer.
Main Methods:
- A systematic literature search was performed using PubMed and Web of Science databases.
- Odds ratios (ORs) and 95% confidence intervals (CIs) were calculated to assess the association between NOD2 polymorphisms and cancer risk.
Main Results:
- The meta-analysis included 30 case-control studies.
- NOD2 rs2066844 C/T, rs2066845 C/G, and rs2066847 (3020insC) polymorphisms were associated with increased cancer risk.
- No significant association was found for the NOD2 rs2066842 C/T polymorphism.
Conclusions:
- NOD2 rs2066844, rs2066845, and rs2066847 polymorphisms may be linked to a higher risk of cancer.
- The rs2066842 polymorphism did not show a significant association with cancer risk.
- Further large-scale studies are recommended to validate these findings.

Cancer-Critical Genes II: Tumor Suppressor Genes
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.
