Blood Studies for Cardiovascular System III: Serum Lipid Profile

854
Understanding serum lipids is crucial for maintaining cardiovascular health and preventing heart disease and stroke.
Serum lipids are fats and fatty substances in the blood and are crucial for various bodily functions, including energy storage, cellular structure, and hormone production. Serum lipids consist of cholesterol, triglycerides, and phospholipids.
Cholesterol is a soft, fat-like substance found in all body cells. It is crucial for producing hormones, vitamin D, and substances that aid...

Area of Science:

  • Clinical Chemistry
  • Lipidology
  • Cardiovascular Disease Risk Management

Background:

  • Accurate blood lipid measurement is essential for managing cardiovascular disease risk.
  • The Centers for Disease Control and Prevention (CDC) Cholesterol Reference Method Laboratory Network (CRMLN) has utilized the beta quantification (BQ) method as a reference measurement procedure (RMP) for high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) for over two decades.
  • Limited data are available on the performance of the BQ RMP.

Purpose of the Study:

  • To assess the long-term performance and accuracy of the beta quantification (BQ) reference measurement procedure (RMP) used by the Japanese Cholesterol Reference Method Laboratory Network (CRMLN) laboratory.
  • To compare lipid measurement results from the CDC lipid reference laboratory and the Japanese CRMLN laboratory.

Main Methods:

  • Comparison of bottom fraction cholesterol (BFC), HDL-C, and LDL-C results obtained via ultracentrifugation.
  • Utilized 280 serum samples analyzed over a 15-year period.
  • Employed statistical method comparison and bias estimation analysis.

Main Results:

  • Regression analysis showed strong agreement between CDC and Osaka laboratories for BFC (R(2)=0.997), HDL-C (R(2)=0.994), and LDL-C (R(2)=0.997).
  • The Osaka CRMLN laboratory consistently met performance goals, aligning with 90% to 95% of CDC reference values.
  • The BQ method demonstrated stability over the 15-year measurement period.

Conclusions:

  • The beta quantification (BQ) method employed by the Osaka CRMLN laboratory is highly accurate and demonstrates sustained performance over more than 15 years.
  • Accurate measurement of bottom fraction cholesterol (BFC) is crucial for precise determination of LDL-C.
  • The findings support the reliability of the BQ method for accurate lipid profiling in cardiovascular risk assessment.
