Regulation of Expression Occurs at Multiple Steps02:24

Regulation of Expression Occurs at Multiple Steps

20.2K
Gene expression can be regulated at almost every step from gene to protein. Transcription is the step that is most commonly regulated. This involves the binding of proteins to short regulatory sequences on the DNA. This association can either promote or inhibit the transcription of a gene associated with the respective sequence.
Transcription results in the generation of precursor (pre-mRNA) that consists of both exons and introns, which needs further processing before being translated to a...

Regulation of Expression at Multiple Steps

1.4K
The gene expression in cells is regulated at different stages: (i) transcription, (ii) RNA processing, (iii) RNA localization, and (iv) translation. Transcriptional regulation is mediated by regulatory proteins such as transcription factors, activators, or repressors—these control gene expression by initiating or inhibiting the transcription of genes. Once a precursor or pre-mRNA is produced, it undergoes post-transcriptional modification, including 5' capping, splicing, and the...

Summary

The basic-helix-loop-helix transcription factor Olig2 is expressed in embryonic cerebellar GABAergic neuron progenitors. This suggests Olig2 plays a role in the early development of Purkinje cells.

Area of Science:

  • Neuroscience
  • Developmental Biology
  • Genetics

Background:

  • The cerebellum contains diverse GABAergic neuron subtypes crucial for its function.
  • Molecular mechanisms driving neuronal subtype diversity are not fully understood.
  • Transcription factors are key regulators of neural development and cell fate determination.

Purpose of the Study:

  • To investigate the expression pattern of the transcription factor Olig2 in the embryonic cerebellum.
  • To determine the role of Olig2 in the development of cerebellar GABAergic neurons, particularly Purkinje cells.

Main Methods:

  • Immunohistochemistry was used to detect Olig2 expression in embryonic mouse cerebella.
  • Co-immunostaining with specific neuronal markers was performed to characterize Olig2-positive cells.

Main Results:

  • Olig2 is expressed in progenitors of GABAergic neurons during peak Purkinje cell generation (embryonic days 11.5-13.5).
  • Olig2-positive cells include both neurogenic progenitors and newly formed neurons.
  • Olig2 co-expresses with Ngn1/2, p57(KIP2), and NeuroD1, but not the early Purkinje cell marker Corl2.

Conclusions:

  • Olig2 is present in early progenitors of cerebellar GABAergic neurons.
  • The expression pattern indicates Olig2 may be involved in the initial stages of Purkinje cell development.
