Six new compounds, attenuatumiones A-F, were isolated from Hypericum attenuatum. Attenuatumione C demonstrated moderate cytotoxic activity against human cancer cell lines SMMC7721 and U2OS.
Area of Science:
- Natural Products Chemistry
- Medicinal Chemistry
- Pharmacognosy
Background:
- Hypericum species are known sources of bioactive compounds.
- Polycyclic polyprenylated acylphloroglucinols (PPAPs) exhibit diverse biological activities.
Purpose of the Study:
- To isolate and characterize new PPAPs from Hypericum attenuatum.
- To evaluate the cytotoxic potential of isolated compounds against human cancer cell lines.
Main Methods:
- Isolation and purification using chromatographic techniques.
- Structure elucidation via comprehensive spectroscopic analysis (NMR, MS).
- Absolute configuration determination using circular dichroism (CD) spectroscopy.
Main Results:
- Six new PPAPs, attenuatumiones A-F (1-6), were identified.
- Twelve known PPAP analogs (7-18) were also isolated.
- Attenuatumione C (3) exhibited moderate cytotoxicity against SMMC7721 and U2OS cells (IC50 values: 10.12 and 10.56 μM, respectively).
Conclusions:
- Hypericum attenuatum is a rich source of structurally diverse PPAPs.
- Attenuatumione C represents a promising lead compound for further anticancer drug development.
