Summary

This study found elevated KRAS oncogene expression in human meningiomas, even without detectable gene mutations. This suggests KRAS may play a role in meningioma development, independent of genetic alterations.

Background:

  • Human meningiomas are tumors arising from the meninges.
  • While chromosomal abnormalities like monosomy 22 are common, specific oncogene involvement is not fully understood.

Purpose of the Study:

  • To investigate the expression of 16 oncogenes in human meningiomas.
  • To determine if KRAS gene expression is altered in meningiomas with normal or monosomy 22 karyotypes.

Main Methods:

  • Dot hybridization and Northern blot analysis were used to detect and quantify oncogene expression.
  • DNA analysis was performed to check for gene amplification or rearrangements in KRAS.

Main Results:

  • KRAS was the only oncogene detected among the 16 investigated.
  • KRAS expression was significantly elevated (6-10 fold) in meningiomas compared to normal meningeal tissue and patient fibroblasts.
  • No gene amplification or major structural rearrangements of the KRAS gene were found.

  • KRAS oncogene is overexpressed in human meningiomas.
  • This overexpression occurs independently of gene amplification or major structural aberrations, suggesting a regulatory mechanism.
  • Elevated KRAS expression may contribute to meningioma development.
