Summary

Interferon therapy can improve hepatitis B e-antigen (HBeAg) seroconversion in chronic hepatitis B patients. However, its effectiveness may be limited by the virus's nuclear DNA, hindering interferon's antiviral activity.

Area of Science:

  • Virology
  • Immunology
  • Hepatology

Context:

  • Chronic hepatitis B virus (HBV) infection is a global health concern.
  • Interferons are known to inhibit HBV replication.
  • Previous studies show improved seroconversion rates with interferon treatment.

Purpose:

  • To evaluate the efficacy of interferon in treating chronic hepatitis B.
  • To investigate the impact of combining interferon with other antivirals.
  • To explore reasons for suboptimal interferon response in chronic hepatitis B patients.

Summary:

  • Interferon therapy significantly increases HBeAg to anti-HBe seroconversion rates in chronic HBV patients compared to no treatment.
  • Combination therapy with interferon and other antivirals did not enhance this effect.
  • Poor therapeutic response may stem from interferon's diminished activity against nuclear HBV-DNA.

Impact:

  • Highlights the potential of interferon as a treatment for chronic hepatitis B.
  • Suggests limitations in current interferon-based treatment strategies.
  • Identifies a potential mechanism (nuclear HBV-DNA) for treatment resistance.
