Purpose of the Study:

  • To investigate the immunogenicity of captopril (CP) in rats.
  • To determine if CP induces specific antibody responses.

Main Methods:

  • Rats received chronic administration of captopril (CP) via intraperitoneal (i.p.) and intramuscular (i.m.) routes.
  • Enzyme-linked immunosorbent assay (ELISA) was used to detect IgG antibodies.
  • Antibody specificity was confirmed through inhibition assays.

Main Results:

  • Chronic CP administration induced a slow-developing IgG antibody response to CP-derived antigens.
  • Antibodies recognized CP-ovalbumin and were inhibited by CP and its derivatives, confirming specificity.
  • A non-drug-specific IgG response against human serum albumin was also observed at high CP doses.

Conclusions:

  • Captopril (CP) is immunogenic in rats, similar to dinitrofluorobenzene but unlike D-penicillamine and benzylpenicillin.
  • The findings suggest that reported anti-CP antibodies in humans may be drug-induced rather than naturally occurring.
